Efficient and gentle delivery of molecules into cells with different elasticity via Progressive Mechanoporation

Description

Progressive Mechanoporation (PM) is a novel mechanoporation method that improves delivery efficiency of molecules into cells of different elasticity. The rational design of our system and the reliable operation and easy-assembly of the PDMS-based chip is optimized to achieve multistage cell deformation through combination of features that provide an improved and unique intracellular delivery by PM. Cas9 protein-sgRNA complex are efficiently delivered high-lighting the biological, biomedical and clinical applicability of our findings.  This represents a further step towards improving mechanoporation irrespectively of cell physical properties and to widen the use of the method.
