Description

Higher electrophoretic mobility DNA (HmDNA) from NaOH-denatured pUC19 reannealed to form native plasmid under renaturing conditions. pUC19 plasmid was denatured using an equal volume of 0.5 N NaOH. After ~5 min, samples were added with hydrochloric acid (HCl) of indicated concentration.  After 10 min samples were analyzed by electrophoresis. Ratio of native plasmid in denatured pUC19 solution was increased with the addition of 0.25 M HCl (lane 4). Experiment was repeated three times (i, ii, iii).

Lane 0, 1 kb ladder; Lane 1, native pUC19; Lane 2, pUC19 + 0.5 N NaOH; Lane 3, pUC19 + 0.5 N NaOH + 0.10 M HCl; Lane 4, pUC19 + 0.5 N NaOH + 0.25 M HCl; Lane 5, pUC19 + 0.5 N NaOH + 0.5 M HCl.
