Description
A new method was established for simultaneous estimation of Ofloxacin and Metronidazole in bulk and dosage form by RP-HPLC method. The chromatographic conditions were successfully developed for the separation of Ofloxacin and Metronidazole by using Qualisil BDS C18 column, flow rate was 1ml/min, mobile phase ratio was Methanol: ACN (70:30%v/v), detection wave length was 254nm. The linearity study for Ofloxacin and Metronidazole was found in concentration range of 10μg-50μg and 10μg-50μg and correlation coefficient (r2) was found to be 0.999 and 0.999, for the amount of drugs estimated by the proposed methods was in good agreement with the label claim. The proposed methods were validated. The accuracy of the methods was assessed by recovery studies at three different levels. Recovery experiments indicated the absence of interference from commonly encountered pharmaceutical additives. The method was found to be precise as indicated by the repeatability analysis, showing %RSD less than 2. All statistical data proves validity of the methods, sensitivity, precision and reproducibility. It can be used for routine analysis of pharmaceutical dosage form. Hence the suggested RP-HPLC method can be used for routine analysis of Ofloxacin and Metronidazole in API and Pharmaceutical dosage form.
