Fig. 8. Substrate specificity of His-tagged, affinity purified recombinant PyPAD1. The activity with p-coumaric acid (8) was arbitrarily set at 100%, and this level of activity represented 2 pkat μg−1 protein. Substrates analyzed are indicated. The enzyme assays were carried out in a buffer containing 100 mM citric acid-Na2HPO4 (pH 3.0) with 50 μM substrates [cinnamic acid (2), p-coumaric acid (8), caffeic acid (9), ferulic acid (10)]. Reactions were performed at 30 °C for 20 min and extracted twice with 100 μl ethyl acetate.
