Metabolomika sa v súčasnosti používa na identifikáciu biomarkerov a zmenených metabolických dráh pri rakovine. Hladiny metabolitov reprezentujú priame molekulárne údaje o stave buniek, ktoré odrážajú zmysluplný fyziologický fenotyp. Cieľom tohto výskumu bolo sledovať metabolomické zmeny v krvi laboratórnych potkanov a vytipovať najdôležitejšie metabolity so zameraním sa na aminokyseliny, biogénne amíny a acylkarnitíny po chemickom navodení nádorov mozgu. V experimente bolo použitých 10 dospelých laboratórnych potkanov kmeňa Sprague Dawley a bolo detegovaných 81 metabolitov. Štatistickými multivariačnými metódami sa ukázali ako významné metabolity acylkarnitíny s krátkym (C3-C5) resp. stredne dlhým reťazcom (C6) a metionín-sulfoxid (Met-SO), a to s ohľadom aj bez ohľadu na pohlavie. Zistené metabolity by mohli predstavovať potenciálne biomarkery, ktoré významne ovplyvňujú diagnostiku a liečbu.
Abstract
Metabolomics is used for the identification of biomarkers and altered metabolic pathways in cancer. The direct molecular data of cell status that display a meaningful physiological phenotype are ultimately represented by the levels of metabolites. The aim of this research was to follow metabolomic changes in the blood of laboratory rats and identify the most important metabolites with a focus on amino acids, biogenic amines, and acylcarnitines after chemical induction of brain tumors. In the experiment were used 10 adult laboratory rats and 81 metabolites were detected. By using statistical multivariate methods, the acylcarnitines with short (C3-C5) or medium-long chains (C6) and methionine sulfoxide were shown to be significant metabolics with the fact that gender was also not considered. Detected metabolites could represent potential biomarkers that significantly influence diagnosis and treatment.
