Supplementary 4: Optimal conditions for Cdc42 siRNA mediated knockdown.

SiRNA mediated knockdown of Cdc42 was evaluated with immunofluorescence for control cells (A) and cells treated with siRNA against Cdc42 (A1). Cdc42 protein was pseudocolored green and DAPI was pseudocolored grey. Cdc42 protein signal was present in cells treated with control siRNA but not in cells treated with Cdc42 siRNA. (B) Immunofluorescent images of control HaCaT cells (B-D), HaCaT cells transfected with universal siRNA (E-G), and Cdc42 siRNA (H-J). DAPI stained the nucleus and was pseudocolored grey (B, E, H). Phalloidin stained F-actin and was pseudocolored green (C, F, I).  Merged channels in a single image were taken for each treatment (D, G, J).
