Here, we provide the custom python code that is used to quantify biophysical parameters of the six dye FluoroCubes and 
all other fluorescent samples used in this study.

The input are .txt files which were saved after processing a TIRF microscopy movie with the Spot Intensity Analysis plugin in ImageJ (https://imagej.net/Spot_Intensity_Analysis). 

The output are plots and they are saved as .svg and .png files.

The following parameters are quantified:
    - Total number of photons
    - Average number of photons per frame
    - Half-life time / bleaching

In addition to the python script we also provide an example data set to test the code (six Cy3 dye FluoroCube) which will recreate some of the plots shown in Figure 1.
