Yildiz Lab FIESTA Adaptations

This repository contains adaptations developed by the Yildiz Lab to the base tracking software FIESTA, originally introduced by Ruhnow et al. (2014)00467-X?_returnURL=https%3A%2F%2Flinkinghub.elsevier.com%2Fretrieve%2Fpii%2FS000634951100467X%3Fshowall%3Dtrue).

Overview

These modifications extend FIESTA's capabilities for high-precision single-molecule tracking and downstream analysis, particularly for MINFLUX-based datasets and step detection workflows.

Key Features

• MINFLUX trace support
  Load and visualize MINFLUX trajectories directly within the GUI framework.

• Interactive step-finding GUI
  Located in the FIONAviewer subfolder, enabling manual and semi-automated step detection.

• Enhanced step analysis tools
  Support for both 1-color and 2-color trajectory analysis, including step size distributions.
  Implemented in the stepping histogram distribution subfolder and integrated with FIONAviewer.
