Remodelling Permissiveness Network (RPN): A Falsifiable Summary

Description
Tissue remodelling capacity is governed by the PAI-1/plasmin/TGF-β fibrinolytic axis. High PAI-1 activity suppresses plasmin, blocking ECM remodelling and locking tissue into a pathological steady state. The RPN framework proposes that therapeutic efficacy — of senolytics, aspirin, and immunostimulatory interventions — is conditioned on baseline permissiveness, not pathology load alone.
