Abstract
Extended-release drug delivery systems are designed to maintain a consistent therapeutic drug
concentration in the bloodstream over an extended period. Duloxetine Hydrochloride is a Selective
Serotonin and Norepinephrine Reuptake Inhibitor (SNRI) widely used in the treatment of major
depressive disorder, generalized anxiety disorder, diabetic neuropathic pain, fibromyalgia, and chronic
musculoskeletal pain. Conventional immediate-release formulations may cause fluctuations in plasma
drug concentration, leading to decreased therapeutic efficacy and potential side effects. Extended-
release formulations provide controlled drug release, minimize peak–trough fluctuations, and enhance
patient compliance by reducing dosing frequency. This review highlights the pharmacological profile
of Duloxetine Hydrochloride, the rationale for extended-release formulations, different formulation
approaches, polymers used in extended-release systems, evaluation parameters, and drug release
kinetics. The review also discusses the future prospects of advanced drug delivery systems for
Duloxetine.
