Genesis Insertion Site Predictor v3: Two Structural Mechanisms Govern Domain Insertion Permissibility

Version 3 of the Genesis Insertion Site Predictor. Expanded validation across 87 experimentally validated domain insertion sites from six structurally unrelated proteins (CAT, MbCas12a, TEM-1, PAC, AraC, SpCas9). Key findings: B-factor z-score achieves mean AUC=0.782 across all six proteins (Binomial test p=0.0016). Discovery of functional dispensability as a second orthogonal mechanism. Combined model AUC=0.881 for enzymes with defined active sites. No GPU or training data required. Web API available at https://app-ebcd5b50.base44.app/functions/predictInsertionSites
