LESE-EIA Structural Coherence Analysis in Solid Oncology: A Cross-Cancer Series — Breast, Colon, Lung, and Pancreatic Ductal Adenocarcinoma
Authors/Creators
Description
We apply the LESE-EIA structural coherence framework (EGESB-G₂ v10.0) to four solid
tumor systems: breast (TCGA-BRCA, n=1,218), colon (TCGA-COAD, n=524), lung
(TCGA-LUAD/LUSC, n=1,070), and pancreatic ductal adenocarcinoma (TCGA-PAAD).
The coherence metric W_ij is derived from the single postulate dW/dτ = κ(1 + log W),
yielding the universal fragility threshold W* = e⁻¹ = 0.36788 without calibration.
Key findings: (1) W_min gradient reproduces the PAM50 prognostic hierarchy in breast
cancer (AUC 0.87, zero calibration); (2) MSI-H/MSS constitutes a structural bifurcation
in colon cancer; (3) LUAD and LUSC represent two structurally distinct W-collapse
trajectories; (4) in PDAC, W* is crossed at PanIN-3 — before invasion and before
clinical detectability — explaining the structural inevitability of late diagnosis.
