Metformin-Rapamycin Combination for mTOR-Mediated Growth Regulation

Description

Metformin activates AMPK to inhibit mTORC1 while rapamycin directly blocks mTORC1 kinase activity, creating synergistic suppression of the hyperactive growth signaling in tetrasomy X. This combination specifically targets the tall stature phenotype by modulating IGF-1/mTOR axis dysregulation caused by increased dosage of X-linked growth-promoting genes.
