Single-cell analysis reveals human-specific oligodendrocyte polarization and conserved neuronal responses after severe traumatic brain injury

Description

This repository contains the analysis pipeline and code for single-cell and single-nucleus RNA sequencing data of human and mouse cortical tissues after severe traumatic brain injury (STBI). The dataset includes samples collected at multiple post-injury time points (<24h, 24-48h, 72-94h for human; 6h, D2, D4 for mouse) to capture dynamic injury responses.The data encompass major brain cell types including microglia, oligodendrocytes, excitatory neurons, enabling cross-species comparisons of transcriptional changes, pseudotime trajectory inference, gene co-expression network analysis, GWAS enrichment, and edema-associated gene correlation studies.Raw and processed sequencing data are publicly available via GEO (accession: GSE277487). Analyses were performed with R (v4.0.5, v4.2.3) and Python (v3.8) using packages such as Seurat, Monocle, hdWGCNA, scVelo, and CellRank.
