Description

MALDI-MSI- meassurements of five murine lungs infected with Aspergillus fumigatus.

Description

This dataset contains MALDI mass spectrometry imaging (MALDI-MSI) measurements of five murine lung tissue samples infected with Aspergillus fumigatus. The data support the manuscript:

Saskia Wolter, Thomas Krüger, Daniela Pelzel, Ilse D Jacobsen, Axel A Brakhage, Ferdinand von Eggeling, Olaf Kniemeyer, Spatial proteomics for the analysis of host-pathogen interactions in mice lungs infected with Aspergillus fumigatusmicroLife, 2026;, uqag010, https://doi.org/10.1093/femsml/uqag010

Biological material and sample preparation

Murine lung tissues from corticosteroid-treated mice experimentally infected with Aspergillus fumigatus (strain CEA17 ΔakuBKU80 ) were analyzed. Tissue samples were formalin-fixed and paraffin-embedded (FFPE), followed by deparaffinization prior to analysis. Sections were cut at a thickness of 5 µm.

For MALDI-MSI analysis, tissue sections were subjected to tryptic enzymatic digestion, followed by application of α-cyano-4-hydroxycinnamic acid (CHCA) as matrix.

MALDI-MSI Acquisition

Measurements were performed using an UltrafleXtreme mass spectrometer (Bruker Daltonics). Data were acquired in a mass range of m/z 500–4000 with a spatial resolution (pixel size) of 50 µm. A medium laser beam size was used with 200 laser shots per pixel.

Data format and File naming convention

Files are provided in standard imzML format, each measurement consists of two files:

  • .imzML file (metadata and spectral coordinates)
  • .ibd file (binary spectral data)

File names follow the structure:
sample_[index]_[internalID]_asp_mm

  • [index]: corresponds to the sample numbering used in the associated publication (ascending order)
  • [internalID]: internal identifier used during data acquisition
