Abstract (English)
Although the „one size fits all” approach is common in the pharmaceutical industry, scientific advances and technologies like 3D printing are increasingly enabling therapies tailored to each patient’s needs. Utilizing the layer-by-layer fabrication process, 3D printing allows control of the parameters important for modification of the drug release profiles. This research aimed to explore 3D printing for developing modified-release dosage forms for oral use and to present examples from the scientific literature and investigational drugs approved by the United States Food and Drug Administration (FDA). The methodology of this paper was based on a narrative review of the scientific literature through a search using the keywords „3D printed modified-release”, „3D printing” and similar. Through literature review, it was found that there are certain parameters that are common to different types of 3D printing technologies. These include the geometric structure of the pharmaceutical form, porosity, infill density, and the choice of excipients. The potential of 3D printing in producing modified-release solid dosage forms, has also been recognized by the FDA, with three investigational 3D-printed candidates from Triastek pharmaceutical company (T19, T20, T21) granted approval for clinical trials. By combining different parameters and various 3D printing technologies, it is possible to achieve modified-release of solid pharmaceutical dosage forms for oral administration. The successful development and clinical trial approval of 3D-printed modified-release formulations highlight the potential of this technology to advance personalized therapies.
