In pharmaceutical new technology, transdermal drug delivery systems (TDDS) have made a fascinating research topic and are one of the most commonly manufactured pharmaceutical products in the global market. These systems can overcome the associated disadvantages of other delivery routes, such as oral and parenteral. A transdermal drug delivery system (TDDS) is a promising delivery system that gives prolonged drug release at a predetermined time. It has many positive consequences, such as passing the first-pass effect, enhancing patient convenience by providing a single application rather than multiple dosing frequencies, and appendageal, intracellular and intercellular. Permeation of drug substances through the stratum corneum remains a significant challenge because it is the rate limiting step for permeation for most molecules. This review article highlights the advances and limitations of the strategies to improve the transdermal delivery of drugs in to the skin, including physical and chemical methods, specifically focusing on providing a comprehensive understanding of transdermal drug delivery systems and enhancement strategies. various penetration enhancing strategies have been successfully employed to enhance the permeation of common and high molecular weight drugs.
