Supplement: NR3C1 is required for normal somatotrope differentiation and Foxo1 expression in pituitary

Description

Supplemental Figure 1. Recombination is evident in the ovary of adult Prop1T2AiCre mice. Immunofluorescence for eYFP (green) and FOXL2 (red) on an adult Stopfl/fl-CAG-eYFP;Prop1T2AiCre ovary section. Recombination is apparent in a proportion of FOXL2-positive granulosa cells. Scale bar represents 100 mm.

Supplemental Figure 2. The number of lactotropes is significantly increased at e16.5 in the absence of glucocorticoid signaling. The number of PRL immunopositive cells was counted in midsagittal sections from Nr3c1fl/fl;Prop1T2AiCre and control embryos at e16.5. Data represent the average ± SEM of counts from three different litters.

Supplemental Figure 3.  Mid-coronal sections from Nr3c1fl/fl;Prop1T2AiCre and control mice at P21 were analyzed by IHC for SOX2 and POMC (green), PRL and GH (red), TSHB and LHB (blue). Nuclei are stained with DAPI (blue). The experiments were replicated twice independently.
