An overview of Alzheimer's disease and therapeutic potential of natural products via In Silico Approach

Alzheimer's disease (AD) is a prominent neurodegenerative illness that causes perpetual loss of intellectual functioning and is a major contributor to dementia cases globally. As per epidemiological statistics, there are approximately 74.7 million AD patients worldwide. The main signs of AD are mostly forgetfulness of recent events and memory loss. In this review we focused on numerous pathways leading to AD, those are oxidative stress, synaptic loss, neuroinflammation, and accumulation of amyloid plaques and neurofibrillary tangles being the major hallmark for the pathology of AD. Natural Product has proved to be best choice of treatment for AD as they offer fewer negative effects and long-lasting advantages. However, many drugs tend to fail clinical trials so as to minimize this possibility computational methods came into light. In this study we discuss the advantages of Computational methods as they are less cost effective, time saving, speeds up the drug design process, requires less animal testing and high success rate using in silico ADMET predictions, molecular docking and dynamic simulation thus making it an excellent tool.  This review summarizes information about Alzheimer disease mechanism and the role of natural products in treatment of AD by using in silico techniques.
