Dataset Overview

This dataset contains raw in vivo two-photon imaging data of axons associated with the publication ‘Impaired axon regeneration and heightened synaptic dynamics in the injured aged mammalian cortex.’ The images were collected from mice as part of the experiments reported in the study and represent the primary data underlying the analyses presented in the paper. Further details on data acquisition and processing are available in the Methods section of the publication.

File Structure

  • Animal-level folders
    Each top-level folder corresponds to one animal. Folder names follow the format:
    Initials–AnimalNumber–Identifier (e.g., CB_005_RTr...).

  • Stitched overview images
    Each folder includes a stitched image showing an overview of axons. This image is created by combining several ROIs, providing context for their spatial arrangement.
    Example: CB_005_RTr_ax1.jpg.
    The star in the stitched image marks where the axon was lesioned.

  • Metadata files
    Each folder contains a .txt file with additional details, including:

    • Imaging coordinates

    • Experimental markers

    • Other acquisition notes

Session and ROI Organization

  • Data is organized into sessions labeled a, b, c, etc.

  • Within each session, files are organized by ROI number.
    Example folder: CB_005_RTr_a-001.

  • If a folder has no session letter, it represents the baseline pre-session image.

Additional Experiments

  • Chronic 2: Contains imaging performed long after the initial experiment to assess axon survival.

  • Degeneration experiments: Stored in separate folders. These track axon degeneration following a lesion over time and use the same folder/session/ROI structure as described above.
