INVESTIGATION OF THE EFFECTS OF PLEUROTUS OSTREATUS, AN EDIBLE MUSHROOM, ON PC-12 ADH CELL DIFFERENTIATION

Pleurotus ostreatus is named as oyster mushroom colloquially and has considerable interest as an edible mushroom around the world. Pleurotus and other members of the Agaricales naturally grow on wood, leaf litter, and/ or other nonwoody lignocellulosic substrates (wood, sawdust, and wheat/rice straw) being used for their commercial production. Recent studies revealed various biological activities including the antibacterial, hypolipidemic, hypoglycemic, antitumor and immunomodulatory properties of exopolysaccharides produced by macrofungus, however, neuritogenic potential of Pleurotus ostreatus exopolysaccharides have not revealed yet. In this study, we aimed to investigate the effects of ethanol extract of Pleurotus ostreatus PC-12 Adh cell differentiation. Because of the neuron-like properties in low serum conditions and with nerve growth factor induction, PC-12 Adh cell line is a very popular in vitro model for studying cell differentiation. MTT assay was performed in order to investigate the cytotoxic effects of Pleurotus ostreatus extract. Subsequently, cells were seeded in low serum conditions and the cell differentiation were evaluated by xCELLigence real time cell analyzer dual plate system according to the changing in cell index values. According to our results, Pleurotus ostreatus extract didn?t show any cytotoxic effects even in high concentrations (IC50>400 ?g/ml) and induced PC-12 Adh cell differentiation significantly with 0.78 ?g/mL concentration according to the nerve growth factor control group. In conclusion, Pleurotus ostreatus, an edible mushroom with cell differentiative properties, might be useful for prevention and treatment of neurodegenerative disorders.
