Antiproliferative and Antimetastatic Effects of Miconazole on Human Non-Small Lung and Colon Cancer Cells

Backgrounds: The rapid development of medicine and pharmacology allows to create effective anticancer drugs. Apparently, there is a need for new therapeutic strategies focusing on inhibition of cancer cell proliferation. The discovery of the antifungal activity of azole compounds represented an important therapeutic advance. Miconazole used as a common treatment for superficial fungal infection. Besides the antifungal activity of miconazole several studies were identified some additional therapeutic effects such as anticancer activity.
Purpose: In this study, was to determine the antiproliferative, anti-migration and anti-invasion effects of miconazole on A549 human non-small lung cancer and Caco2 colon cancer cells.
Methods: To evaluate anticancer potential of the miconazole, the methods measuring various antiproliferative endpoints were employed including MTT assay, real time monitoring of cell proliferation, anti-invasion and anti-migration with Real time cell analyzer system. The cytotoxic effects of different concentrations were investigated on both cells and then IC50 values were calculated.
Results: Cell proliferation assay was performed with miconazole IC50 concentrations (28.5 ?g/mL for Caco2 and 31.5 ?g/mL for A549 cells). Subsequently, antimetastatic efect were conducted on cancer cell lines with the IC50 concentrations. Miconazole was effected on cell death and antimetastatic activities of both cancer cells. Furthermore miconazole was significantly inhibited invasion and migration activities of A549 compared to Caco2 cells.
Conclusions: This study should be supported by molecular cell mechanisms of antitumor effects of miconazole for lung and colon cancer therapies.
