Raw data and processed data of resolution dependence on the average STED beam power in a STED holographic endoscope

Description

Raw data of imaged 400nm and 500nm fluorescent nanospheres for assessing the Full Width at Half Maximum (FWHM) dependence on the STED average beam power from Figure 2. To be used with the Supplementary Code: '100sphere_analysis.py'.
The ZIP files 400nm.zip and 500nm.zip contain folders with the respective raw data. 

Each folder contains HDF5 files with the following name structure:

m[number]_p[power].hdf5 - STED images (excitation + depletion beam at a specific power)
m[number]_p[power]_b.hdf5 -  Background images (no excitation, just depletion beam at a specific power) [to subtract to the respective image above)

[number] - from 0 to 9 | correspond to the image number
[power] - values from the following list (0,119,123,126,131,137,140,143,147,153). Note that these values do not correspond to the real power values!
The real average power values can be seen in the file FWHM1-10_data_400nm_final.hdf5 or FWHM1-10_data_500nm_final.hdf5, respectively, under the dataset name 'Intensity'.

Total of 10 images numbers, at 10 depletion power each (100 measurements x 2 sphere sizes [400nm and 500nm]), with respective background (except for 0mW depletion beam power). Each image was done always in brand new sphere (no sphere was reused to avoid influence of photobleaching).

The processed data from Figure 2 can be found in the files FWHM1-10_data_400nm_final.hdf5 and FWHM1-10_data_500nm_final.hdf5, for 400nm and 500nm nanospheres, respectively. These datasets were obtained by processing the previous files using the Supplementary Code: '100sphere_analysis.py'. The files contain 5 datasets:
Average FWHM - average full width at half maximum over 10 measurements/images at a specific STED depletion beam power
Average StandardDeviationstandard deviation of full width at half maximum of 10 measurements/images at a specific STED depletion beam power
FWHM - full width at half maximum for all images spheres (10 spheres at 10 depletion powers each)
Intensity - STED depletion beam average power for each measured/imaged sphere
Intensity StandardDeviation - standard deviation of the intensity values (power fluctuation)

The result of the fit model from Figure 2 is available in the file StedFit.h5. The file has 4 datasets:
ism - intensity
fwhm0_29 - full width at half maximum extrapolated for a 0nm sphere, corresponding to the effective STED point spread functino
fwhm400_29n - full width at half maximum fit for the 400nm nanosphere datapoints
fwhm500_29n - full width at half maximum fit for the 500nm nanosphere datapoints
