Plasmid Maps of DUF247 Constructs

We have generated full-length DUF247 constructs, as well as three fragments - fragment A (from amino acid number 1 to 216), fragment B (from amino acid number 217  to 468) and fragment C (from amino acid number 469 to 503). Fragments A, B, AB, and ABC (full length DUF248) were cloned into entry vector pGGC (GreenGate) with mVenus as N-terminal tag. DUF247 fragments were PCR amplified using high-fidelity DNA polymerase, and cloned as full-length cDNA or truncated version. The entry clone was evaluated using Sanger sequencing to verify the absence of mutations. Promoters used included Ubiquitin 10 promoter.
