Raw NGS Data for "Mapping the Functional Landscape of the Receptor Binding Domain of T7 Bacteriophage by Deep Mutational Scanning"

This directory contains relevant fastq files used for DMS analysis in the publication “Mapping the Functional Landscape of the Receptor Binding Domain of T7 Bacteriophage by Deep Mutational Scanning”. Note if one is looking for functional scoring for each variant, a complete summary of functional scoring on each host is available in a dramatically more convenient excel file attached as Source Data for Figure 2 and Figure 4 in the publication.

Fastq files for each replicate are already split with the name of the folder or file noting the host used for selection and number of infection cycles or using an L code which indicates the following:

The data analysis performed uses the scripts located here https://github.com/raman-lab/oracle, and more explicit instructions can be found in the methods section of the publication. Data can be further split by internal barcodes as noted in the paper for technical replicates.

Also note we have since updated the scripts we use for this kind of analysis (they are now better, faster, stronger etc.) so if you have come here to compare notes on that regard feel free to reach out and we can provide more recent guidance.
