A nonenzymatic dependency on inositol-requiring enzyme 1 controls cancer cell cycle progression and tumor growth
Figure 2A: AMO1 shIRE1 cl.1 (left) or shXBP1 cl.1 (center) cells were incubated in the absence (dark 215 grey) or presence (light grey) of Dox (0.2 μg/ml) for 72 h. Cells were stained with propidium iodide (PI) and analyzed by flow cytometry to determine cell cycle phases by DNA content. Sub-G1, G0/G1, S, and G2/M peaks are annotated for the shIRE1 histograms. Representative plot of 3 independent experiments.
Figure 2D: Flow cytometry histograms representing AMO1 shIRE1 cell proliferation after 12 (left) or 48 (right) hour culture in the presence or absence of Dox (0.2 μg/ml) measured by CFSE staining. Generations following cell division are color coded and represented in the schematics on the right. Representative plot of 3 independent experiments.
