    "description": "BACKGROUND: Exposure to allogeneic blood products\noften leads to the development of human leukocyte\nantigen (HLA) antibodies. Refractoriness to platelet\n(PLT) transfusion caused by alloimmunization against\nHLA Class I antigens constitutes a significant clinical\nproblem.\nSTUDY DESIGN AND METHODS: We developed an\nRNA interference (RNAi)-based approach to silence the\nexpression of HLA Class I molecules on PLTs derived\nfrom CD34+ progenitor cells. A lentiviral-based system\nwas used to express short-hairpin RNA (shRNA) targeting\nb2-microglobulin (b2m) transcripts in CD34+ progenitor\ncells. Differentiation to PLTs was performed by\nincubating progenitor cells in the presence of thrombopoietin\nand interleukin-3.\nRESULTS: The transduction of RNAi cassettes containing\nthe sequences for shRNAs targeting b2m caused\nup to 85% reduction of progenitor cells HLA Class I\nantigen expression, which was maintained in the\nculture-derived PLTs. The HLA-deficient PLTs derived\nfrom HLA-silenced CD34+ cells proved to be fully functional\nin in vitro tests when compared to peripheral\nblood\u2013derived PLTs.\nCONCLUSIONS: Our data show that in vitro generating\nHLA Class I\u2013deficient PLTs from hematopoietic progenitor\ncells prove to be feasible. As malignancy risks associated\nwith insertional mutagenesis are not to be\nexpected in anucleated PLTs, provision of HLA-deficient\nPLTs from large-scale production units may become\nreality in the management of patients suffering from\nPLT transfusion refractoriness.",
