General Information
This dataset involves analysis of mice 3 month after injection of alpha-synuclein monomers.
Brain sections were stained with antibodies against alpha-synuclein, VGLUT1, and VGLUT2.
Analysis of co-localization of VGLUT1 or VGLUT2 with alpha-synuclein was performed.


Data Specific Information
This file contains the percentage of VGLUT1 or VGLUT2 puncta on an imagewise basis that have been identified as colocalized with alpha-synuclein.

Methodology
This data was collected using a Zeiss 900 Airyscan microscope with a 63x 1.4NA objective.
Images were Airyscan processed in Zeiss Zen Blue with a strength setting of 5.5.
Background subtraction was performed in Imaris, and the Coloc function was applied to quantify colocalization of VGLUT1 with α-synuclein and, in a separate analysis, VGLUT2 with α-synuclein. 
Threshold parameters were determined independently for each channel using the automatic thresholding function (calculate threshold). 
Colocalization was then assessed using the Build Coloc Channel function in Imaris, and the percentage of VGLUT1- or VGLUT2-positive voxels above threshold colocalized with α-synuclein were determined. 
Statistical analysis was performed using a linear mixed-effects model.
